If you are using seasons I am 85% sure the herbicide will still work yes. It has been a while since I had to try it though, I tend to have all my fertilizing done before plants germinate so it isn't an issue.
You don't need to spray the entire field, just run a test strip down the middle of something. Farmers run test strips for experiments in real life all the time.
Why would you have to start a new game? I have stuff not working out the way I thought it would all the time, I just adapt and keep going.
